What is BioTrap 500?

BioTrap 500 is a new biocompatible sample extraction column, enabling repeated direct injections of plasma, serum, urine, milk, supernatants of cell cultures, fermentation broth and other complex matrices into the HPLC-system, without previous clean-up procedures (except for a simple centrifugation).

Prior to HPLC analysis, macromolecular compounds such as proteins, must be removed. Otherwise, the high concentrations of organic modifiers normally used in the mobile phases in reversed- phase chromatography, will cause precipitation of the proteins. By using BioTrap 500, proteins and other macromolecules can be totally removed (on-line) from the sample, before analysis of the drug molecules on the analytical column.

The BioTrap 500 column is a silica based extraction column with a biocompatible external surface and a hydrophobic internal surface (C18 or C8 groups). The biocompatibility has been obtained by attachment of the plasma protein alpha1-acid glycoprotein (AGP) on the external surface of the particles. AGP is an extremely stable protein which tolerates organic solvents used in reversed-phase HPLC, without being denaturated. The pores of the particles are small enough to exclude proteins and other macromolecular compounds, which means that they can be directly flushed to waste. Small drug molecules, however, can penetrate the pores of the particles and be adsorbed to the C18/C8 groups on the inner surface.

Repeated injections of up to 1000 µl plasma can be made directly onto the BioTrap 500 column (20 x 4.0 mm I.D.). By using optimal conditions, a total amount of more than 100 ml of plasma may be injected onto the BioTrap 500 column.
